Bioactivity Comments
In calcium mobilisation and β-arrestin recruitment assays PF-06767832 shows high cooperativity with acetylcholine (ACh) to activate the M1 receptor, but exhibits little intrinsic agonist activity [2]. PF-06767832 increases efficacy of ACh by 126-fold in the calcium assay, and 316-fold in the β-arrestin assay. This is coupled with ~2.5-fold bias toward β-arrestin over G-protein mediated calcium signalling.
