Comment: Netrin 1 is a 604 amino acid peptide. Structurally it contains laminin EGF-like domains, a netrin domain, and a C terminal domain that acts as a cell attachment site. In normal cells netrins are involved in regulating cell guidance, particularly during embryonic development. Netrin 1 is overexpressed in some cancers, and has been associated with tumourigenesis. Experimental evidence suggests that netrin 1 promotes epithelial-to-mesenchymal transition (EMT) and metastasis [2]. Conversely, disrupting netrin 1 (expression or function) has anti-tumour effects [1-2].
Receptors for netrin 1 include DCC netrin 1 receptor (DCC) and unc-5 netrin receptor B (UNC5B); unliganded these receptors promote apoptosis, but in the presence of netrin 1 they induce an EMT.
