Comment: ASP7663 is a selective activator of the TRPA1 ion channel [1]. It activates human, rat, and mouse channels with similar EC50 values in vitro, and is effective by oral administration in rodent models of drug-induced constipation and abdominal pain.

Selectivity for TRPA1 was confirmed by screening against more than 60 receptors, including channels (like TRPV1) and enzymes [1] (see Supplemental table S1 and Supplemental figure S1 therein for full details). Results demonstrated that ASP7663 had no or only slight affinity for any of these tested protein targets. |
