Comment: Tepotinib is a potent and selective, orally available inhibitor of MET tyrosine kinase [1]. It was designed to inhibit the pro-oncogenic signalling caused by MET gene alterations that occur in 3-5% of NSCLC cases, and which correlate with poor prognosis.

This compound induces regression of human tumours in murine xenograft models, and appears to be well tolerated [1]. In a kinase screening panel only 5 of the other 241 kinases were inhibited by 10μM tepotinib (EMD 1214063) by more than 50%; IRAK4, TrkA, Axl, IRAK1, and Mer [1]. |
