Programmed necrosis (or necroptosis), is induced by TNFα and is dependent upon RIPK1 action [1]. RIPK1-dependent necroptosis is a key driver of the pathogenesis of inflammation and disease, for example in ischemia/reperfusion injury in the brain [8], retina [5] and kidney [3], and in models of myocardial infarction [6] and retinal detachment [7]. RIPK1 inhibitors would help redude such TNFα-induced pathologies.
