protriptyline is an approved drug (FDA (1967))
Compound class:
Synthetic organic
Comment: Protriptyline is a tricyclic antidepressant.
Marketed formulations may contain protriptyline hydrochloride (PubChem CID 6603149). Ligand Activity Visualisation ChartsThese are box plot that provide a unique visualisation, summarising all the activity data for a ligand taken from ChEMBL and GtoPdb across multiple targets and species. Click on a plot to see the median, interquartile range, low and high data points. A value of zero indicates that no data are available. A separate chart is created for each target, and where possible the algorithm tries to merge ChEMBL and GtoPdb targets by matching them on name and UniProt accession, for each available species. However, please note that inconsistency in naming of targets may lead to data for the same target being reported across multiple charts. ✖ |

Summary of Clinical Use |
Used to treat major depressive disorder and ADHD, sometimes used to treat narcolepsy. |
Mechanism Of Action and Pharmacodynamic Effects |
Tricyclic antidepressants inhibit reuptake of serotonin and norepinephrine by their respective transporters, SERT (SLC6A4) and NET (SLC6A2), and in addition anatgonise many neurotransmitter receptors. Inhibition of SERT and NET increases synaptic neurotransmitter concentrations which enhances neurotransmission [1,4]. Antagonism of receptor activation may have clinical efficacy but may also contribute to the drug's side effects. See the DrugBank link for further details. |
