Thiethylperazine is used to relieve nausea and vomiting. Marketing authorisation for this drug has been discontinued in the US, but other national approval agencies may continue to grant marketing authorisation.

Thiethylperazine is a dopamine antagonist. The antiemetic effect may be associated with central antidopaminergic actions, with potential activity within the medullary chemoreceptor trigger zone (controls the vomiting center). A novel drug action is reported by Krohn et al (2011). This study shows that thiethylperazine-induced activation of the transport protein Abcc1 (an ATP-binding cassette transporter aka Mrp1), in a mouse model of Alzheimer's disease (AD) reduces the cerebral amyloid-β (Aβ) load [1].
