Comment: Thymic stromal lymphopoietin (TLSP), an IL-7 like cytokine, is the endogenous ligand for the TLSP receptor. TLSP/TLSPR signalling is pro-inflammatory.

TLSP/TLSPR signalling promotes release of pro-inflammatory Th2 mediators by mast cells and eosinophils [4], enhances eosinophil chemotaxis and pro-survival properties [8], and modulates mast cell proliferation and differentiation from progenitors [3]. TLSP is elevated in patients with severe asthma [7] and atopic dermatitis [6]. Anti-TSLP agents offer potential for the treatment of a variety of inflammatory conditions [1]. For example, the anti-TLSP mAb, tezepelumab (a.k.a. AMG 157 or MEDI9929), has completed Phase 1 evaluation in atopic dermatitis [2] (see NCT00757042) and Phase 2 for asthma (NCT02054130).
