Comment: Tegoprubart (AT-1501) is a monoclonal antibody that targets human CD40 ligand (CD40LG; CD154). Signalling via the CD40-CD40L costimulatory pathway is a recognized mechanism that can drive transplant (graft) rejection and inhibition/blockade of this pathway is considered as a potential therapeutic approach to achieve transplant tolerance and graft survival [3-5]. Tegoprubart is modified to prevent FcγRII receptor-mediated platelet activation and reduce the risk of drug-induced thromboembolic events.
