Comment: Manfidokimab (AK120) is a humanized IgG4 monoclonal antibody that targets the IL-4 receptor alpha (IL-4Rα) subunit that is a component of the IL-4 and IL-13 receptor complexes. It was designed to disrupt signaling mediated by IL-4 and IL-13, in particular to block the pathological T helper 2 (Th2)-mediated inflammatory responses initiated by these cytokines. Peptide sequences for the heavy and light chain domains of manfidokimab match those that form humanized antibody 13E5 H2L2 as claimed in Akeso Pharmaceuticals' patent US20230295312A1 [1].
