Comment: EG-2184 is a pannexin 1 channel (Px1) blocker [1]. Its structure was developed from that of probenecid, and it is one of the claims in Aphiotx patent WO2023283743A1 [2]. Pannexin 1 channels have been associated with noradrenergic activity and neuronal hyperexcitability in the locus coeruleus during opioid withdrawal. Blocking channel activity is proposed as a pharamcological mechanism to counteract the physical and aversive symptoms of opioid withdrawal syndrome.
