Comment: GC-373 is the parent of the dipeptide-based prodrug GC-376. The aldehyde drug and bisulphite adduct prodrug were originally reported as inhibitors of the 3CL protease (Mpro) of feline coronaviruses and caliciviruses [1]. They also inhibit Mpro of both SARS-CoV and SARS-CoV-2, and inhibit SARS-CoV-2 replication in cell culture [3].
