Comments: PVRIG has been shown to interact with nectin-2 (a.k.a. PVRL2: NECTIN2; Q92692) [1]. This interaction mediates a co-inhibitory signal that blocks T cell activation/proliferation [1,3].
Immunopharmacology Comments
PVRIG has been identified as a novel co-inhibitory immune checkpoint, that is exploited by some cancer cells to evade immune detection [1-2]. Disruption of the PVRIG/PVRL2 interaction by antagonistic anti-PVRIG antibodies reduces tumour growth in preclinical models [1]. An anti-human PVRIG monoclonal antibody called COM701 is being developed by Compugen for immuno-oncology potential in patients with advanced solid tumours- see Phase 1 clinical trial NCT03667716 which is evaluating COM701 as a monotherapy and in combination with the PD-1 checkpoint inhibitor nivolumab.
