Immunopharmacology Comments
CD147 might induce the production of pro-inflammatory cytokines such as IL-6 in monocytes [1]. Meplazumab (Jiangsu Pacific Meinuoke Bio Pharmaceutical) is a clinical stage anti-CD147 monoclonal that is being repurposed for COVID-19 pneumonia (caused by the coronavirus SARS-CoV-2). In this setting it has potential to both inhibit viral entry into host cells via CD147, and to block CD147-mediated inflammatory signalling.
General Comments
Basigin (CD147) is a multi-functional immunoglobulin superfamily protein that is widely expressed (e.g. in epithelial cells, endothelial cells and leukocytes). It has metalloproteinase-inducing ability, and regulates biological processes including spermatogenesis, expression of the monocarboxylate transporter and the responsiveness of lymphocytes. Endogenous ligands include cyclophilins [2] and some integrins.

Basigin is exploited by the merozoite stage of Plasmodium falciparum to invade erythrocytes. CD147 neutralising antibodies can block merozoite invasion of host erythrocytes [3], indcating the potential of basigin as a novel malarial drug target in the host. Meplazumab (Jiangsu Pacific Meinuoke Bio Pharmaceutical) is a clinical stage anti-CD147 monoclonal that is being investigated for potential as both prophylaxis and treatment of malaria.
