PDB Ligand Immunopharmacology Ligand
Compound class: Synthetic organic
Comment: Compound 41 is a structurally novel, potent, selective and orally bioavailable PI3Kδ inhibitor [1]. Results from this inhibitor optimisation study suggest that off-target inhibition of Vps34 is associated with in vivo toxicity in a rat PK model.

Immunopharmacology Comments
Given the leukocyte-restricted expression of PI3Kδ this kinase has been targeted for the development of anti-inflammatory pharmaceuticals for the treatment of respiratory diseases, chronic obstructive pulmonary disease, autoimmune disorders and chronic inflammatory diseases. Isoform selectivity is an important element of drug development, to prevent off-target side-effects at the ubiquitously expressed α and β isoforms, particularly with respect to the long-term treatment required for chronic diseases.
