The DYRK1A gene is located within the Down Syndrome (DS) critical region on chromosome 21. Trisomy of chromosome 21 leads to overexpression of DYRK1A and amyloid precursor protein (APP), and this may underlie the Tau and amyloid pathologies associated with early onset Alzheimer's disease in DS [1-3,5]. Pharmacological inhibition of DYRK1A activity would aim to modify the course of AD pathogenesis.
