Comment: Compound 18c is a calcium-sensing receptor (CaSR) negative allosteric modulator (NAM) [2]. It utilises a second allosteric binding site that is topographically distinct from the NPS2143 binding site [1]. Negative modulation of CaSR promotes release of parathyroid hormone (PTH). Transient release of PTH elicits a bone anabolic response, so molecules that act as rapid/transient negative CaSR modulators are being explored for potential as novel agents to treat bone-loss diseases.
